In general, the understanding of the pathogenesis, diagnosis, and treatment of CMPN has been repeatedly revised over the long history of its study. In recent years, significant progress has been made in establishing the role of the JAK/STAT signaling pathway in Ph-negative CMPN. This has led to the identification of molecular targets for directed pathogenetic therapy with targeted drugs capable of modifying the course of these diseases and significantly increasing the duration and quality of life of patients, and has also allowed the creation of an international unified system of criteria for diagnosis, monitoring and assessment of response to treatment. The continuous integration of molecular diagnostics, new drugs targeting STATs, and combination therapies is necessary to improve therapeutic accuracy and clinical outcomes. Future studies should offer a deeper understanding of the influence of the basic mechanisms of the JAK/STAT signaling pathway on the development of the disease, which should maximize the effectiveness of the use of Janus kinase inhibitor drugs and minimize side effects in patients at different stages of these diseases.
